PE2228 is a synthetic, seven–aminoacid peptide (sequence GVSWGLR), designed as a shortened analog of spadin and a potent antagonist of the TREK1 twopore potassium channel (IC₅₀ ≈ 0.12 nM) By blocking TREK1—expressed in brain regions tied to mood, learning, and memory—it produces rapid antidepressant effects, inducing hippocampal neurogenesis and synaptogenesis in just a few days. Animal studies indicate it alleviates depressive-like behavior in ~4 days, boosts markers like BDNF and CREB, and improves cognitive function with minimal side effects.  It also shows promise in stroke recovery and may support neuroprotection and muscle contractility via TREK1 modulation. In sum, PE2228 represents a fast-acting, nootropic and neuroregenerative spadin analog under active preclinical research.
